Case Sharing for EmCD CHO® 121 Series
● Support scale-up of the CHO-S cell process
Experimental objective: To address issues such as rapid cell death, decreased viability, and low expression titer during scale-up of the CHO-S cell process. This experiment evaluates the impact of EmCD CHO® 121 medium series and competitor media combinations on cell viability, cell growth, and target product expression by simulating shake flask and 3-L bioreactor conditions using a fed-batch process.
Experimental results: Under the premise of consistent quality, the expression titer with the EmCD CHO® 121 medium series during scale-up were significantly superior to that with the competitor media combinations. This demonstrates that EmCD CHO® 121 medium series can more effectively support CHO-S cell growth and promote the expression of the target product.
A data comparison of the culture performance between EmCD CHO® 121 medium series and competitor media combinations (shake flask and 3-L bioreactor) is as follows:
Compared to competitor media combinations, EmCD CHO® 121 medium series exhibited a more stable yield during the fed-batch process scale-up of CHO-S cells. In contrast, the yield of competitor media combinations in the bioreactor process showed a rapid decline (by 33%).
