CHO cells are currently the most common expression system used in the production of recombinant biopharmaceuticals. Continuously upgrading and optimizing culture medium formulations, Eminence introduces the brand-new CHO cell media-EmCD CHO® 121 medium series. The EmCD CHO® 121 medium series are chemically defined, devoid of hydrolysates, proteins, growth factors and other substances. The EmCD CHO® 121 series comprises the Basal Medium and Feeds A/B.

The EmCD CHO® 121 medium series display strong versatility, are suitable for CHO-S, CHO-K1, CHOZN, and CHO-DG44 cells, and can maintain high cell viability and yield. CHO-S cells are favored by process development technicians for their rapid growth rate. However, issues such as rapid cell apoptosis and failure to maintain the viability during process scale-up often occur, leading to low expression titer and certain quality concerns. EmCD CHO® 121 is a high-performance medium series specifically developed for CHO-S cell lines to effectively address the issues during the scale-up process, including rapid cell death, decreased viability and low expression titer. It enables cells to maintain high density and viability in bioreactor systems, thereby enhancing the protein yield and quality.

Case Sharing for EmCD CHO® 121 Series

Support scale-up of the CHO-S cell process

Experimental objective: To address issues such as rapid cell death, decreased viability, and low expression titer during scale-up of the CHO-S cell process. This experiment evaluates the impact of EmCD CHO® 121 medium series and competitor media combinations on cell viability, cell growth, and target product expression by simulating shake flask and 3-L bioreactor conditions using a fed-batch process.

Experimental results: Under the premise of consistent quality, the expression titer with the EmCD CHO® 121 medium series during scale-up were significantly superior to that with the competitor media combinations. This demonstrates that EmCD CHO® 121 medium series can more effectively support CHO-S cell growth and promote the expression of the target product.

A data comparison of the culture performance between EmCD CHO® 121 medium series and competitor media combinations (shake flask and 3-L bioreactor) is as follows:

图片6

Compared to competitor media combinations, EmCD CHO® 121 medium series exhibited a more stable yield during the fed-batch process scale-up of CHO-S cells. In contrast, the yield of competitor media combinations in the bioreactor process showed a rapid decline (by 33%).

图片7

The EmCD CHO® 121 medium series exhibited comparable cell growth and maintained higher cell densities compared to competitor media combinations in shake flask and bioreactor.

图片8

The EmCD CHO® 121 medium series showed comparable cell growth and maintained better cell viability compared to competitor medium combinations in shake flask and bioreactor.

图片9

The EmCD CHO® 121 medium series had lower mannose content (Man5 <2%) compared to competitor media combinations in shake flask and bioreactor.

图片10

The EmCD CHO® 121 medium series showed comparable critical quality attributes (CEX) to competitor media combinations, with lower acidic peaks (<20%) and higher main peaks (>70%).

图片11

The EmCD CHO® 121 medium series exhibited comparable critical quality attributes (SEC) to competitor media combinations, with a main peak reaching 97% and containing lower fragments and aggregates (<3%).
